SFC Platform for the Purification of Compounds

The Kromasil SFC platform of chromatographic columns for Supercritical Fluid Chromatography (SFC) is based on first-in-class Kromasil silica, and supplied in a variety of column formats, for quick analysis and seamless scale-up. Kromasil SFC columns support users worldwide in research, discovery and compound isolation of small molecules, amino acids and peptides.

SFC is an alternative for separating and purifying substances and is regarded as green chemistry. Its advantage is being a low-cost technique using carbon dioxide as eluent with small amounts of organic solvents as modifier. By using CO2, with a CapEx investment for CO2 recycling, it can be a fairly sustainable separation method with regard to mobile phase consumption.

Whether in analytical or semi-prep scale, 2.5 or 5 μm particles, Kromasil offers the same material produced in a similar way with similar specifications. This means users can be sure that the separation itself will stay identical during scale up. All Kromasil SFC phases are available in columns up to 30 mm i.d. In this way we can offer products that give users the possibility to seamlessly transition between different particle and column sizes and easily scale up their separations.

Kromasil SFC XT, the result of an inorganic merged silica technology is also included in the Kromasil SFC portfolio. SFC XT offers separations mechanism that are different to traditional SFC stationary phases. Because of the high surface availability of the SFC line, our materials show high loadability which makes it excellent choices for purification under overloaded conditions. The figure illustrates an example of a scale up and purification of prednisolone crude.

In similarity to LC, almost all preparative separation starts at analytical scale using smaller particles and/or smaller columns. By developing the preparative method on an analytical scale, SFC users can minimize solvent and sample usage while shorten time for method development.
